Elastic Fiber EVG Stain
Intended use:
Elastic fiber EVG stain is useful in demonstrating hyperplasia, fracture, rupture, or fragmentation of elastic tissue as an aid of diagnosis. Observe collagen fiber and muscle fiber via counterstaing with VG Solution.
Principle:
The principle of this method is not well-known. Hypothesis includes that hydrogen bond forms between certain parts of elastic fiber and phenolic group of resorcin in Elastin Stain, consequently the elastic fiber is stained blue-black. VG stain is counterstaining.
Methods:
1. Deparaffinize and hydrate sections to water.
2. Oxidate in Potassium permanganate for 5 minutes. Rinse in distilled water.
3. Bleach in Oxalic acid for 5 minutes. Rinse in distilled water.
4. Rinse slightly in 95% ethanol and stain in Elastin Stain for 8~24 hours.
5. Differentiate in 95% ethanol or 1% hydrochloric acid (observe under the microscope if necessary).
6. Rinse thoroughly in tap water and distilled water.
7. Counterstain in VG Solution for 1 minute. Differentiate quickly in 95% ethanol for several seconds. Dehydrate in absolute ethanol and clear in xylene. Mount with mounting media.

Precaution:
1. Differentiate tissue directly in 95% ethanol after Elastin Stain, without rinsing in water.
2. As elastin Stain is relatively volatile and takes a long time to complete the staining, dip and stain in sealed container.
3. After staining in VG solution, do not rinse in water but directly drop 95% ethanol to differentiate and dehydrate in absolute ethanol. Otherwise, the colors stained with VG solution will weaken or even discolor.
