Acid Phosphatase (ACP) Stain

Intended Use:
This kit is for staining bone marrow cell and bloodcell smear.

 

Principle:
Acid Phosphatese stain is based on  diazotization-coupling principle.  At pH0.5, Acid Phosphatase hydrolyzes Naphthol AS-Bl Phosphoric Acid and releases Naphthol AS-B1 to mix with diazonium salt and form insolublecolored precipitate in cytoplasm.

Methods:


Working Solution Preparation (for 1 test only)


Apparatus required: disposable tube, micropipette, disposable tips, dropper


Instruction: Mix 25µl solution B with 25µl solution C; wait for 2 minutes. Add distilled water 2ml into 25µl solution D and 90µl solution E. Mix thoroughly.

 

Procedure:

1. Dry the smear and add solution A to fix the smear for 30 seconds. Rinse the smear with distilled water and

    dry it.

2. Add working solution and incubate at 37℃for 60 minutes. Rinse with distilled water and dry it.

3. Counterstain with solution F for 1~2 minutes. Rinsewith distilled water. Dry the smear for microscopic

    examination.
